The synergy of this kit allows for the study of intracellular coenzyme levels from two distinct vectors. While exogenous NAD⁺ provides the direct substrate for sirtuin activation and ATP production, 5-Amino-1MQ acts as a membrane-permeable inhibitor of the enzyme Nicotinamide N-methyltransferase (NNMT). Research suggests that by blocking NNMT, 5-Amino-1MQ prevents the degradation of nicotinamide, theoretically preserving the endogenous pool of NAD⁺.
